ABSTRACT

To investigate the herbal prescription rules of Professor Jiang Liangduo in the treatment of abdominal mass based on the traditional Chinese medicine inheritance support system software (TCMISS) of version 2.5, find out new herbal formulas for the treatment of abdominal mass, and then provide new reference to its traditional Chinese medicine therapy. By the method of retrospective study, one hundred and thirty-two outpatient prescriptions of Professor Jiang for the treatment of abdominal mass were collected to establish a typical database with TCMISS. Four properties, five tastes, channel tropism, frequency count, Chinese herbal prescriptions rules and the new prescriptions were analyzed so as to dig out the prescription rules. There were 57 herbs with a frequency>=15, and then 91 core combinations of 2-5 herbs were evolved and 9 new prescriptions were created. It was found out that these drugs mainly had the effects of liver nourishing and soothing, soft-moist and dredging-tonifying, supporting right and dispeling evil, cooperating with the method of calming the liver and resolving hard lump according to the actual situation. It reflected the thought of treatment based on syndrome differentiation in TCM, and provided a new reference for its clinical treatment and research.

ABSTRACT

Objective To develop a rat model of periapical periodontitis caused by repeated root canal infection with Enterococcus faecalis (E. faecalis), and to evaluate the disease progression by observing the periapical lesion area, inflammation, and TNF-a expression in the root apex. Methods Thirty Sprague Dawley rats were used in this study. The pulps of the bilateral first maxillary molar teeth of each rat were exposed, absorbent cotton with lipopolysaccharides (LPS) was implanted, and then the teeth were exposed in the oral environment for four weeks to induce periapical periodontitis of primary root canal infection with mixed bacteria. Then the root canal was subjected to disinfection treatment for two weeks, followed by inoculation with E. faecalis suspension for one week to produce repeated root canal infection model with E. faecalis for a 3-week observation. X-ray, H-E staining and immunohistochemical staining were used to observe the periapical lesion area, inflammation, and TNF-a expression in the root apex at one week, two weeks, three weeks after establishment of repeated root canal infection model; the above parameters were also observed for periapical periodontitis caused by primary root canal infection with mixed bacteria (6 weeks later) and for root canal disinfected by Ca (OH) (for 6 weeks). Results (1) Periapical periodontitis caused by repeated root canal infection with E. faecalis had gradually increased periapical lesion area, with unclear damage border; the infection reached a severe level during the 1-2 weeks and became chronic inflammation at week three; and the expression of TNF-a began to decrease after two weeks. (2) The periapical periodontitis caused by primary root canal infection with mixed bacteria had a smaller periapical lesion area, with clear damage border, slighter infiltration of inflammatory cells, and less TNF-a expression. (3) The periapical periodontitis with root canal disinfected by Ca (OH)2 had the smallest periapical lesion area, with disappeared inflammation, slight TNF-a expression, and hyperplasia of cementum and alveolar bone. Conclusion E. faecalis has a strong periapical damaging effect, with continuous bone absorption observed even under the chronic inflammatory status.

ABSTRACT

The aim of the present study is to investigate the effects of adiponectin (APN) on hypoxia/reoxygenation (H/R) injury in cultured cardiomyocytes. Primary cardiomyocytes were obtained from neonatal rats by enzymatic digestion method and identified by immunofluorescent technique. Primary cells cultured for 72 h were used in experiment and divided into 5 groups randomly: Control group, H/R group, H/R+APN group, H/R+APN+adenine 9-beta-D-arabinfuranoside (AraA, AMPK inhibitor) group, and H/R+AraA group. The cardiocyte morphology and beating rate were observed under inverted microscope. The DNA ladder was examined by agarose gel electrophoresis, and the cell apoptosis was determined by flow cytometry. Moreover, the malondialchehyche (MDA) content in myocardial cells and the superoxide dismutase (SOD) activity in the supernatant were measured using kits, the fluorescence intensity of intracellular Ca2+ was observed by laser scanning confocal microscope, and the phosphorylation of AMPK was determined by Western blotting. Compared with control group, H/R group showed increased apoptotic rate, oxidative stress level, intracellular Ca2+ concentration and phosphorylation level of AMPK (P<0.05), while significant ameliorations in the above indices were seen in H/R+APN group. On the contrast, AraA attenuated the protective effect of APN and decreased the phosphorylation of AMPK. These results suggest that adiponectin can protect cardiomyocytes from H/R-induced oxidative stress and apoptosis through AMPK pathway.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the AvaII polymorphism of low density lipoprotein receptor gene in both health and essential hypertension populations, and to evaluate the association of AvaII polymorphism with level of blood lipid.</p><p><b>METHODS</b>Using polymerase chain reaction (PCR), AvaII polymorphism was studied in 109 health individuals and 319 patients with essential hypertension.</p><p><b>RESULTS</b>There were three kinds of genotype: (+/+), (+/-), (-/-). The frequencies of the three genotypes were shown as follows: (+/+) 0.9%, (+/-) 27.5%, (-/-) 71.6% in health population and (+/+) 1.2%, (+/-) 33.9%, (-/-) 64.9% in essential hypertension population, respectively. The frequencies of the two alleles were shown as follows: (+) 14.7%, (-) 85.3% in health population, (+) 18.2%, (-) 81.8% in essential hypertension population and (+) 17.3%, (-) 82.7% in the community, respectively. In male essential hypertension cases, the genotypes were associated with serum TC and LDL-C level in the following order: (-/-) < (+/-) (P < 0.05). In women and healthy population, there was a similar trend but not statistically significant (P > 0.05).</p><p><b>CONCLUSIONS</b>A significant association was found between the common variation of LDL-R gene and serum TC and LDL-C levels. (+) Allele was associated with elevated level of serum TC and LDL-C, but (-) allele was associated with a low level of serum TC and LDL-C. The frequencies of (-) allele in both group were related to serum low level TC while LDL-C was much higher than that reported in the western countries. These data indicated that genetic factors which resistant to hypercholesterolemia in Chinese people were different from those findings in West while might be one of the reasons to explain why that serum TC level in Chinese was lower than people in the western countries.</p>
